Resistance mutation in 60% of cases
Four lead compounds were identified with predicted binding affinity below -10.0 kcal/mol against the EGFR T790M gatekeeper mutation. Osimertinib derivative structures showed favourable ADMET profiles with predicted blood-brain barrier permeability, a critical property for central nervous system metastases. The Lazertinib analogue demonstrated the highest CNS penetration score. All four candidates passed the Lipinski filter with zero violations. Next steps involve in vitro kinase assays and cellular viability testing against H1975 cell lines.
